Short answer
About a week with a loading phase, or about a month without one. That is how long studies took to fill muscle with creatine: 20 grams a day raised stores about 20% in 6 days, and 3 grams a day did the same in 28 days. Short trials found performance gains after 5 days of loading. Early effects without loading are less certain.

How fast muscle stores fill
In a 1996 study in the Journal of Applied Physiology, 31 men took creatine on different schedules. Muscle creatine rose about 20% after 6 days at 20 grams a day. At 3 grams a day, it rose by a similar 20%, but gradually, over 28 days.
The International Society of Sports Nutrition's 2017 position stand puts it the same way: 3 to 5 grams a day fills muscle over 3 to 4 weeks. On a normal diet with 1 to 2 grams a day from food, it says muscle stores are about 60 to 80% full, which is the gap supplements close.
When performance changes showed up
A 1993 trial gave 12 people either creatine (5 grams four times a day) or a glucose placebo for 5 days. After creatine, peak muscle force during repeated bouts of all-out exercise was higher; placebo showed no change. The ISSN says performance in high-intensity, repeated exercise generally rises 10 to 20% after loading.
Without loading, the evidence is thinner. In a 2011 trial, about 2.3 grams a day for 6 weeks made 20 young adults more resistant to fatigue, with no change in maximal strength or body weight.
Why you might not notice anything
These trials measured force on machines during repeated, hard efforts, not how people felt day to day. Starting point matters too: in the 1992 study that first showed loading works, the rise in muscle creatine was greatest in people who started with low levels, so those already near the top have less room to gain.
Related questions
Does taking more make creatine work faster?
Loading fills muscle faster, but not higher: 20 grams a day for 6 days and 3 grams a day for 28 days reached a similar level in the 1996 study.
How long does creatine stay in your muscles after you stop?
In the 1996 study, levels were back to baseline about 30 days after stopping. The ISSN says it generally takes 4 to 6 weeks.
